What is Benlysta?

Benlysta (belimumab) is a monoclonal antibody and immunosuppressant that belongs to the class of medications called BLyS-specific inhibitors. It is only approved for use as an add-on to standard therapy to treat active lupus nephritis, progressive multifocal leukoencephalopathy (PML), severe active central nervous system lupus, and active systemic lupus erythematosus (SLE) in both children and adult patients. Benlysta can be administered in two ways: either once a week as a subcutaneous (SC) self-injection or once a month (after the initial doses) as an intravenous (IV) injection administered by a medical expert. Frequent adverse effects include fever, diarrhea, and nausea.

Important information about Benlysta

The active ingredient in Benlysta is belimumab, which is a biologic medicine. It is considered a B-lymphocyte stimulator (BLyS)-specific inhibitor. The mechanism of action involves inhibiting the function of a protein known as BLyS to reduce your immune system's B cell activity. This may lessen inflammation and disease activity.

Follow all directions given to you by your doctor and read all documents provided, including the full Prescribing Information, FDA-Approved Product Labeling, Instructions for Use, and Medication Guide, before using Benlysta. However, the following is a synopsis of the general guidelines for the use of Benlysta:

  • A healthcare professional may administer Benlysta intravenously (IV infusion) or it can be given subcutaneously (subcutaneous administration), either by self-administration or by a healthcare professional.
  • It is possible to prescribe Benlysta as a single-dose prefilled syringe or as a single-dose autoinjector.

The most common side effects of Benlysta seen in clinical trials include:

  • Diarrhea.
  • Fever.
  • Infusion reactions.
  • Nausea.
  • Bronchitis.
  • Depression.
  • Sore throat.
  • Bladder infection or urinary tract infection (UTI).
  • Pain in the arms or legs.
  • Trouble sleeping.

Some individuals may experience serious side effects while using Benlysta. These side effects may require immediate medical attention. If you experience any of the following, contact your healthcare provider or seek emergency medical help:

  • Confusion, fever, dyspnoea, weakness, coughing, perspiration, chills, stomach discomfort, diarrhea, and burning when urinating are all signs of serious infections.
  • Breathing difficulties, hives, itching, a rapid heartbeat, tightness in the throat, nausea, stomach pain, lightheadedness, vertigo, and swelling in the face or tongue are all signs of a serious, life-threatening allergic reaction.
  • Depressive symptoms, suicidal thoughts, and self-harming ideas or actions.

These are not all of the possible side effects of Benlysta. You should always seek medical advice from your healthcare provider for any questions or concerns about your medical condition or treatment. Read all patient information, medication guides, or drug information sheets that come with this medication. You can also report adverse effects to the U.S. Food and Drug Administration at www.fda.gov/medwatch or 1-800-FDA-1088.

Before starting Benlysta, it is crucial to discuss any existing health conditions with your healthcare provider. You should be sure that your healthcare provider is aware of all your medical conditions, including if you have:

  • An active infection.
  • Worsening feeling of depression and suicidal thoughts.
  • A weakened immune system.
  • Have recently had a live vaccine or have one scheduled.

Patients who have experienced belimumab-induced anaphylaxis should not take Benlysta, as well as those who have hypersensitivity reactions or adverse reactions to Benlysta. You should also avoid this medication if you are under 5 years of age or are also using other biologic therapies or cyclophosphamide.

The speed at which Benlysta (belimumab) worked was not the main focus of clinical studies. Instead, they examined whether lupus nephritis or systemic lupus erythematosus (SLE) got better or worse over time. To find out how Benlysta is working for you, you may need to take this drug for one to two years, depending on your condition. To keep an eye on your health throughout this time, you will have routine examinations and blood testing. If you are unsure of what to anticipate during Benlysta treatment, ask your healthcare team.

When Benlysta is taken with other prescription drugs, over-the-counter medications, vitamins, and supplements, it may change how they work or increase the frequency or severity of side effects. Make sure that you tell your healthcare professional about anything that you are taking to avoid any negative drug interactions, including:

  • Rituximab.
  • Azathioprine.
  • Corticosteroids.

The condition for which you are receiving Benlysta (belimumab), the method of injection (subcutaneous or intravenous), your age, and your body weight will all be taken into consideration by your healthcare team when determining your dosage.

Subcutaneous Injection (SC):

18 years of age and up:

SLE: Once a week, 200mg injections are usually injected beneath the skin.

400 mg administered subcutaneously once weekly for four doses is the usual dosage for lupus nephritis. Following four doses, the weekly dosage will be 200 mg.
