Important information about Parsidol

The active ingredient in Parsidol is ethopropazine hydrochloride (HCl). Its chemical structure (c19h24n2s) is related to 10h-phenothiazine-10-ethanamine structures and has strong anticholinergic activity. Because its structure contains a diethylamino side chain related to n-diethyl substitutions found in similar phenothiazines, it behaves like other small molecules within this class.

It is a tricyclic phenothiazine derivative with anticholinergic, parasympatholytic, and antispasmodic effects. It reduces the effects of acetylcholine in the central nervous system (also called an acetylcholinesterase - AChe inhibitor or butyrylcholinesterase - BChe inhibitor) by blocking muscarinic receptors. This helps restore balance between dopamine and cholinergic pathways in patients with Parkinson's disease. It is considered a profenamine-type compound and is structurally related to profenamine hydrochloride. It also has weak antihistamine properties.

Follow all directions given to you by your doctor and read all documents provided, including the full Prescribing Information, FDA-Approved Product Labeling, Instructions for Use, and Medication Guide before using Parsidol. However, the following is a synopsis of the general guidelines for the use of Parsidol:

  • Parsidol tablets are taken by mouth.
  • If you miss a dose, take it when remembered unless it is close to the next dose. Skip the missed dose if necessary.
  • Do not stop suddenly; dosage should be reduced gradually.

Some mild or common side effects of Parsidol seen in clinical trials as compared to placebo include:

  • Drowsiness or tiredness.
  • Dizziness or mild headache.
  • Dry mouth.
  • Stomach pain.

Some individuals may experience serious side effects while using Parsidol. These side effects may require immediate medical attention. If you experience any of the following, contact your healthcare provider or seek emergency medical help:

  • Tachycardia (fast heart rate) or orthostatic hypotension (feeling dizzy when standing up suddenly, especially with large initial doses.
  • Rare worsening of Parkinsonism or ataxia.
  • CNS stimulation with high doses, including irritability.
  • Cholestatic jaundice or skin reactions.
  • Symptoms of an allergic reaction, such as a skin rash.

These are not all of the possible side effects of Parsidol. You should always seek medical advice from your healthcare provider for any questions or concerns about your medical condition or treatment. Read all patient information, medication guides, or drug information sheets that come with this medication. You can also report adverse effects to the Food and Drug Administration at www.fda.gov/medwatch or 1-800-FDA-1088.

Before starting Parsidol, it is crucial to discuss any existing health conditions with your healthcare provider. You should be sure that your healthcare provider is aware of all your medical conditions, including:

Prostatic hypertrophy or risk of urinary retention.

Pyloric stenosis.

Cardiovascular disease.

Parsidol should not be used in:

Patients with glaucoma.

Those with hypersensitivity to phenothiazines.

In clinical use, anticholinergic antiparkinsonian medications typically show benefit within days as dose adjustments are made.

When Parsidol is taken with other prescription drugs, over-the-counter medications, vitamins, and supplements, it may change how they work or increase the frequency or severity of side effects. Make sure that you tell your healthcare professional about anything that you are taking to avoid any negative drug interactions, including:

  • Anticholinergic drugs such as aclinidium or inhaled anticholinergics should be avoided in combination with Parsidol as they may increase the side effects or toxicity of it.
  • Drugs such as clozapine, eluxadoline, and pramlintide may increase the risk of constipation when used with Parsidol.
  • Anticholinergic agents (like ethopropazine) increase the tachycardic effects of dronabinol; the combination should be avoided.
  • Potassium salts can lead to ulcers when used with Parsidol.

Tablets containing 50 mg ethopropazine hydrochloride.

It should be stored at room temperature out of reach of children or pets.

Parsidol is no longer on the market, but Parsitan is an alternative trade name manufactured by Searchlight Pharma Inc.

Yes, it is available from many manufacturers of generic drugs, and under the brand name Parkin.
