Important information about Prolopa

The active ingredients in Prolopa are levodopa and benserazide. Symptoms of Parkinson’s disease are linked to low dopamine in the brain. Levodopa helps replace dopamine, while benserazide helps more levodopa reach the brain and may reduce some levodopa-related side effects outside the brain.

Follow all directions given to you by your doctor and read all documents provided, including the full Prescribing Information, FDA-Approved Product Labeling, Instructions for Use, and Medication Guide before using Prolopa. However, the following is a synopsis of the general guidelines for the use of Prolopa:

  • Take Prolopa by mouth in divided doses.
  • Swallow capsules whole with water. Do not open the capsules or dissolve them in liquid.
  • When starting Prolopa, the dose is increased slowly. It may take several weeks to find the best dose.
  • Taking Prolopa with or right after a low-protein snack, such as fruit, applesauce, or biscuits, may help reduce stomach side effects.
  • Protein-rich meals may reduce how well Prolopa works.
  • If you miss a dose, take it as soon as you remember. If it is almost time for your next dose, skip the missed dose and return to your regular schedule.
  • If you have severe Parkinson's symptoms, do not resume usual activities quickly once your symptoms start to improve, as you may be at risk of injuring yourself.
  • This medication can make you drowsy, so don't plan to drive or operate any heavy machinery until you know how it affects you.

Some mild or common side effects of Prolopa include:

  • Dry mouth.
  • Headache.
  • Sleepiness or drowsiness.
  • Sweating.
  • Rash or itching.
  • Dark colour in sweat or urine.

Some individuals may experience serious side effects while using Prolopa. These side effects may require immediate medical attention. If you experience any of the following, contact your healthcare provider or seek emergency medical help:

  • Abnormal involuntary movements, such as twitching, shakiness, spasms, or movements that may look like Parkinson’s symptoms.
  • Hallucinations, confusion, depression, anxiety, agitation, nightmares, or other mental changes.
  • Allergic reaction, including hives, itching, swelling of the lips, face, tongue, or throat, or trouble breathing or swallowing.
  • Irregular heartbeat or palpitations.
  • Light-headedness when rising quickly from a sitting position.
  • Excessive sleepiness or suddenly falling asleep without warning.
  • Impulse control symptoms, such as compulsive gambling, spending, eating, or increased sexual urges.
  • Signs of dopamine dysregulation syndrome include excessive use of the medication beyond the recommended dose.
  • Signs of neuroleptic malignant syndrome, which include fever, changes in heart rate or blood pressure, muscle stiffness, and altered mental status.
  • Skin changes.

These are not all of the possible side effects of Prolopa. You should always seek medical advice from your healthcare provider for any questions or concerns about your medical condition or treatment. Read all patient information, medication guides, or drug information sheets that come with this medication. You can also report adverse effects to the Food and Drug Administration at www.fda.gov/medwatch or 1-800-FDA-1088.

  • Before starting Prolopa, it is crucial to discuss any existing health conditions with your healthcare provider. You should be sure that your healthcare provider is aware of all your medical conditions, including:
  • Osteoporosis.
  • History of heart problems, including heart attack or irregular heartbeat.
  • Diabetes.
  • History of peptic ulcer.
  • Management of intention tremor, Huntington’s chorea, or drug-induced extrapyramidal effects. Prolopa is not indicated for these conditions.
  • Wide-angle glaucoma or uncontrolled intraocular pressure.
  • Upcoming surgery with halothane.
  • History of skin cancer (melanoma) or undiagnosed skin lesions.

  • Contraindication to sympathomimetic amines such as epinephrine, norepinephrine, or isoproterenol.
  • Within two weeks of a monoamine oxidase inhibitor (MAOI).
  • Severe heart disease.
  • Severe endocrine, kidney, liver, or lung disease.
  • Known allergy or hypersensitivity to levodopa or benserazide, as well as inactive ingredients in the formulation, such as titanium dioxide.
  • Pregnant or breastfeeding patients.

The monograph says the best dose may take several weeks to reach. For many patients, optimum dosage can usually be reached within 2 to 3 weeks, but response varies by person.

When Prolopa is taken with other prescription drugs, over-the-counter medications, vitamins, and supplements, it may change how they work or increase the frequency or severity of side effects. Make sure that you tell your healthcare professional about anything that you are taking to avoid any negative drug interactions, including:

  • Monoamine oxidase inhibitors - may cause serious unwanted effects, including hypertensive crises.
  • Sympathomimetic drugs, such as epinephrine, isoproterenol, norepinephrine, or amphetamine - levodopa may increase their effects.
  • Blood pressure-lowering medications - postural low blood pressure may occur, so blood pressure may need monitoring.
  • Reserpine-containing medications may reduce the action of Prolopa.
  • Antipsychotics with dopamine-blocking effects may reduce Prolopa’s benefit or worsen Parkinson’s symptoms.
  • Psychoactive drugs may increase the risk of unusual side effects and need careful monitoring.
  • Halothane anesthesia may cause blood pressure changes or arrhythmias during surgery.
  • Iron tablets or multivitamins with iron may lower levodopa levels in the body.
  • Isoniazid, phenytoin, papaverine, metoclopramide, domperidone, opioids, and other antiparkinsonian medications - may change Prolopa’s effect or increase side effects.
  • Protein-rich diets may reduce the beneficial effects of Prolopa.

Prolopa is available as an oral capsule dosage form in 3 strengths:

  • 50 mg levodopa/12.5 mg benserazide.
  • 100 mg levodopa/25 mg benserazide.
  • 200 mg levodopa/50 mg benserazide.

It should be stored at room temperature, out of the reach of children and pets.

Prolopa is manufactured by Hoffmann-La Roche Limited.

No, but both active ingredients are available as single-entity products.

Do not stop taking Prolopa suddenly unless your doctor tells you to. If Prolopa needs to be reduced or stopped, your doctor should taper it slowly and monitor you for serious symptoms. If therapy is interrupted for a long period, a lower dose may be restarted and adjusted gradually.

Prolopa is contraindicated during pregnancy and in those of childbearing potential unless adequate contraception is used. If pregnancy occurs while taking Prolopa, it must be discontinued by tapering as advised by the prescribing physician.

It is not known whether benserazide passes into breast milk. Those who require Prolopa should not breastfeed because skeletal effects in infants cannot be excluded.

Talk to your doctor if you are already breastfeeding or if you have any plans to breastfeed. They will know or recommend what is best for you.
