Important information about Ortho Tri-Cyclen
Tri-Sprintec is a combination hormonal birth control that contains ethinyl estradiol, an estrogen, and norgestimate, a progestin. It works mainly by preventing ovulation and also changes cervical mucus and the uterine lining, making it harder for sperm to reach an egg and for pregnancy to occur.
Follow all directions given to you by your doctor and read all documents provided, including the full Prescribing Information, FDA-Approved Product Labeling, Instructions for Use, and Medication Guide before using Tri-Sprintec. However, the following is a synopsis of the general guidelines for the use of Tri-Sprintec:
- Tri-Sprintec is taken once daily at the same time each day.
- Tri-Sprintec is a triphasic oral contraceptive with active tablets in different strengths - light blue, blue, and white “active” pills, followed by green placebo tablets. Following the order of the package, take one active tablet daily for 21 days, followed by 7 days of placebo tablets.
- Start taking the first active tablet as directed by your healthcare provider.
- If you miss one active tablet, take it as soon as you remember and then take the next tablet at your regular time (this may mean taking two tablets in one day).
- If you miss two active tablets in a row in the first or second week, take two tablets on the day you remember and two tablets the next day, then continue taking one tablet daily. Use a backup method of contraception, such as spermicide or condoms, for at least 7 days.
- If you miss two active tablets in a row in the third week or miss three or more active tablets at any time, skip the rest of the active tablets and start a new pack. Use a backup method of contraception for at least 7 days.
- If you miss any placebo tablets, discard the missed tablets and continue taking one tablet daily until the pack is finished.
- Tri-Sprintec does not protect you against sexually transmitted diseases. Continue using barrier contraception during sexual activity to prevent transmission.
Some mild or common side effects of Tri-Sprintec seen in clinical trials as compared to placebo include:
- Nausea.
- Breast tenderness or breast pain.
- Headache.
- Bloating.
- Weight gain.
- Breakthrough bleeding or vaginal bleeding between menstrual periods.
- Contact lens intolerance.
Some individuals may experience serious side effects while using Tri-Sprintec. These side effects may require immediate medical attention. If you experience any of the following, contact your healthcare provider or seek emergency medical help:
- Signs of a heart attack include chest pain, shortness of breath, and pain radiating to your shoulder, neck, or jaw.
- Signs of a stroke include numbness on one side of your body, a severe headache, or trouble speaking.
- Signs of a blood clot/ thromboembolism (deep vein thrombosis or pulmonary embolism), including severe leg cramps, swelling or redness, chest pain, or fast heart rate.
- Signs of cancer include breast lumps/ pain, swollen lymph nodes, or unusual vaginal bleeding.
- Signs of gallbladder disease include pain or tenderness in your stomach.
- Severe allergic reaction presenting with swelling of the face, lips, tongue, or throat, hives, and trouble breathing.

Before starting Tri-Sprintec, it is crucial to discuss any existing health conditions with your healthcare provider. You should be sure that your healthcare provider is aware of all your medical conditions, including:
- A history of blood clots, thrombosis, or thromboembolic disorders.
- Heart valve problems or coronary artery disease.
- High blood pressure (hypertension), high cholesterol, or high triglycerides.

When taken correctly, Tri-Sprintec begins working as a method of contraception within the first cycle. Backup contraception may be needed during the first 7 days, depending on when you start.
When Tri-Sprintec is taken with other prescription drugs, over-the-counter medications, vitamins, and supplements, it may change how they work or increase the frequency or severity of side effects. Make sure that you tell your healthcare professional about anything that you are taking to avoid any negative drug interactions, including:
- Carbamazepine, phenytoin, grapefruit juice, rifampin, St. John’s wort, and other enzyme-inducing drugs may reduce the effectiveness of hormonal contraceptives.
- Ritonavir and other protease inhibitors may affect hormone levels.
- Thyroid hormones may require dose adjustments.
- Ombitasvir, paritaprevir, ritonavir, and dasabuvir may increase liver enzyme levels.

- Gray tablets- 0.18 mg/0.035 mg norgestimate/ ethinyl estradiol.
- Light blue tablets- 0.215 mg/0.035 mg norgestimate/ ethinyl estradiolare light blue.
- Blue tablets - 0.25 mg/0.035 mg norgestimate/ ethinyl estradiol.
- Tri-Sprintec 28 also contains 7 white placebo tablets (no active component).
