Qinlock
Qinlock (ripretinib) is a kinase inhibitor that is indicated to treat adult patients with advanced gastrointestinal stromal tumors (GIST). GIST is a cancer that mostly exists in the stomach and intestines but can happen anywhere in the gastrointestinal tract. You must have received treatment for your advanced GIST previously with at least three kinase inhibitors, including imatinib, sunitinib, and regorafenib, to receive treatment with Qinlock.

Important information about Qinlock
How does Qinlock work?
The active ingredient in Qinlock is ripretinib. Qinlock is an enzyme inhibitor and is part of the drug class of cancer medicines known as tyrosine kinase inhibitors. Its mechanism of action is to block the activity of receptors called KIT and PDGFRA on the surface of cancer cells. By stopping the enzyme, the use of Qinlock will stop cancer cells from growing and multiplying. This helps improve your progression-free survival (PFS), or the time from initial treatment to growth of the cancer or death.
How do you use Qinlock?
Follow all directions given to you by your doctor and read all documents provided, including the full Prescribing Information, FDA-Approved Product Labeling, Instructions for Use, and Medication Guide before using Qinlock. However, the following is a synopsis of the general guidelines for the use of Qinlock:
- Take one tablet at the same time daily
- You can take your medication with or without food
- Swallow the tablets whole; do not chew, crush, or split the tablets
- Do not take an extra dose if you vomit your normal dose. Instead, speak to your healthcare provider
- Do not change your dosage unless your healthcare provider tells you to do so

Some individuals may experience serious side effects while using Qinlock. These side effects may require immediate medical attention. If you experience any of the following, contact your healthcare provider or seek emergency medical help:

- New primary cutaneous malignancies, including:
- Skin cancers known as cutaneous squamous cell carcinoma, keratoacanthoma, or melanoma.

- Tell your doctor right away if you experience any symptoms of cardiac failure, such as shortness of breath, swelling in your abdomen, or you’re feeling very tired.
- Risk of impaired wound healing
- Photosensitivity
- Let your healthcare provider know if you have any skin changes.
- Wear sunscreen whenever possible; Qinlock will make you burn more easily.
This is not the full list of adverse reactions, however, these were the ones found in the Invictus study. Invictus was a global, randomized, double‑blind, placebo‑controlled Phase 3 trial in patients who had an unresectable, locally advanced, or metastatic gastrointestinal stromal tumor (GIST), and who had been treated with at least three prior treatments. The primary efficacy outcome measure was progression-free survival (PFS). The secondary efficacy outcome was the overall survival rate. Talk to your oncology team about any concerns you may have.
You should always seek medical advice from your healthcare provider for any questions or concerns about your medical condition or treatment. Read all patient information, medication guides, or drug information sheets that come with this medication. You can also report adverse effects to the Food and Drug Administration at www.fda.gov/medwatch or 1-800-FDA-1088.
